Actividad antitrombótica de proteínas de amaranto

Con el fin de obtener conocimientos que sirvan de aporte para futuros desarrollos de alimentos funcionales que contengan a las proteínas de amaranto como uno de sus ingredientes, en este trabajo nos hemos propuesto estudiar los aspectos básicos de la actividad antitrombótica de las proteínas de amaranto. Se estudiaron diversos protocolos de hidrólisis sobre los aislados proteicos de amaranto como una herramienta que nos permitiese generar distintos polipéptidos y péptidos activos. En todos los casos se observó que el tratamiento enzimático sobre las proteínas de amaranto generó péptidos con potencial actividad antitrombótica, siendo mucho más efectivo cuando la hidrólisis fue realizada por las enzimas gastrointestinales utilizadas en la simulación. Los resultados prometedores obtenidos con el aislado sometido a la digestión gastrointestinal nos impulsaron a seleccionarlo para ahondar en la búsqueda de péptidos con actividad antitrombótica presentes en las proteínas de amaranto, realizándose determinaciones in vitro, in vivo y ex vivo sobre la muestra. En los ensayos realizados con animales se pudieron estudiar parámetros asociados a las distintas etapas de la hemostasia, sugiriendo los resultados que el aislado proteico de amaranto es un potencial agente antitrombótico. Los péptidos que se encuentran encriptados en las proteínas de amaranto, una vez liberados durante la digestión gastrointestinal serían capaces de ejercer un efecto antiplaquetario y/o anticoagulante. Con el objeto de intentar comprobar la hipótesis anteriormente mencionada, se buscó purificar aquellos péptidos responsables de la actividad antitrombótica y estudiar la potencial biodisponibilidad de los mismos. Se logró, de este modo, obtener una fracción con muy alta actividad antitrombótica in vitro y las pruebas de absorción realizadas utilizando esta fracción activa, junto con su secuenciación nos permitieron encontrar péptidos potencialmente bioactivos, algunos de los cuales lograron atravesar el epitelio intestinal representado por la monocapa celular desarrollada en el inserto. Los resultados encontrados a lo largo de este trabajo de tesis fueron consistentes y nos inducen a pensar que en las proteínas de amaranto se encuentran encriptados péptidos bioactivos capaces de ejercer una actividad antitrombótica. Queda aún pendiente esclarecer el o los mecanismos de acción involucrados.

The aim of the Journal is to provide a forum for the critical analysis of advanced drug and gene delivery systems and their applications in human and veterinary medicine. The Journal has a broad scope, covering the key issues for effective drug and gene delivery, from administration to site-specific delivery.

In general, the Journal publishes review articles in a Theme Issue format. Each Theme Issue provides a comprehensive and critical examination of current and emerging research on the design and development of advanced drug and gene delivery systems and their application to experimental and clinical therapeutics. The goal is to illustrate the pivotal role of a multidisciplinary approach to modern drug delivery, encompassing the application of sound biological and physicochemical principles to the engineering of drug delivery systems to meet the therapeutic need at hand.

Articles review the current status of a specific topic, giving equal emphasis to the identification of major conceptual and technological challenges to successful drug and gene delivery, to an evaluation of triumphs as well as shortcomings in current conceptual and technical approaches, and to a discussion of their possible solution.

Theme issues are commissioned by one of the Executive Editors or the Editor-in-Chief. The Journal does not publish stand alone manuscripts. If you wish to submit a theme issue topic for consideration, please contact one of the editors.

Marine Biotechnology is a scientifically and economically expanding enterprise that is poised to harness the enormous but uncharted functional diversity of marine life, with its novel and rich array biodesigns and biosynthetic capabilities. From this pursuit comes new genes, chemicals, materials and inspirations for the benefit of industry, nutrition, medicine and the sustainable use and management of the world’s oceans. This Special Issue in Marine Drugs highlights the cutting-edge developments in Marine Biotechnology with a collection of papers written by authors who are leading experts in the field including selected papers from the 10th International Marine Biotechnology Conference (IMBC-2013), the premier meeting in marine biotechnology under the auspices of the International Marine Biotechnology Association.
