The objective of this thesis was to evaluate the angiogenic process and its possible mechanism of regulation in the pregnant rat uterus and cervix. In addition, investigate the influence of neonatal exposure to xenoestrogenic substances on the development of the uterine vascular compartment and in the female reproductive performance. Results: 1) Using intact pregnant rats, we have demonstrated that the vascular tree of the uterine cervix is an active cellular compartment showing striking changes in the relative area occupied by vessels, endothelial proliferation and vascular maturation. These modifications were associated with changes in vascular endothelial growth factor (VEGF) expression. 2) The mast cells degranulation could be a necessary process to control the normal angiogenesis of the rat cervix during pregnancy and we suggest that the effect occur via a VEGF dependent pathway. 3) Neonatal exposure to xenoestrogen altered the normal steroid hormone-responsiveness in the uterus of adult female rats, showing a fail in VEGF gene activation and a decrease in endothelial cell proliferation. In parallel, changes in estrogen and progesterone receptors expression were found at a time where an abnormal overexpression of the corepressor SMRT was showed, suggesting that neonatal xenoestrogen exposure alters the transcriptional machinery of steroid-dependent genes. 4) Finally, in addition to mentioned effects, females exposed to xenoestrogens were subfertile. The results obtained with this Thesis aspire to contribute to the understanding of angiogenesis as a necessary physiological process for a successful labor. In addition, provide experimental evidences about the effects of xenoestrogen exposure on reproductive health.Palabras clave – provistas por el repositorio digital
