Péptidos bioactivos producidos por bacterias lácticas con actividad inhibitoria del desarrollo de microorganismos

Lactobacillus plantarum LP31 strain, isolated from Argentinian dry-fermented sausages, produced an bactericidal substance active against some spoilage and foodborne pathogenic bacteria. It was preliminarily characterized as a bacteriocin and was sequencially purified by solid phase extraction, gel filtration chromatography and reverse phase. We have found that this bacteriocin is a peptide with approximately fourteen aminoacids and have molecular weight of 1558.85 Da. Plantaricin-149 is a bacteriocin produced by Lactobacillus plantarum NRIC. In this work, a synthetic peptide analog denoted as Pln149a and a hybrid peptide (HP) with the N-terminal amino acid sequence os Pediocin PA-1 was prepared by solid phase. The antagonistic activity against gram-positive and gram-negative bacteria was tested, and the secondary structure was studied by Circular Dichroism (CD) and Fluorescence Spectroscopy, under several conditions. Plna was inhibitory against Staphylococcus aureus and Listeria strains. The HP and its C-terminal part, was active only against Listeria strains, in accordance with the results found for Pln149a. Analysis of secondary structure, found that Pln149a acts via the carpet mechanism. According to the results obtained with the hybrid bacteriocin we conclude that the molecule contains two domains: a cationic, N-terminal β-sheet domain that mediates binding of the bacteriocin to the target cell surface and an amphipathic C-terminal α-helical domain that possibly may be involved in the penetration into the hydrophobic part of the target cell membrane.
