Construcción y evaluación de virus MVA recombinantes que expresan la glicoproteína D del virus BoHV-1

El virus vaccinia Ankara modificado (MVA) es un virus altamente atenuado que se utiliza eficientemente como vector viral no replicativo para el desarrollo de nuevas vacunas. En este trabajo de Tesis se desarrolló un nuevo inmunógeno basado en MVA que expresa como antígeno de interés la glicoproteína D (versión secretada, gDs) del virus herpes bovino tipo I (BoHV-1), un agente infeccioso ampliamente distribuido en Argentina. Primeramente, se diseñó y construyó el vector de transferencia para obtener MVA recombinantes utilizando como sitio blanco de inserción el gen no esencial MVA086R. Se obtuvieron virus MVA-gDs recombinantes y se demostró la correcta expresión, antigenicidad y N-glicosilación de la glicoproteína D expresada a partir de dichos virus. Posteriormente, se evaluó la capacidad inmunogénica de los virus MVA-gDs en animales de experimentación (ratones y conejos). En el modelo de ratón, se observó que los virus MVA-gDs desencadenaron una respuesta inmune específica, de tipo humoral, que se mantuvo en niveles similares durante un período de siete meses (luego de la aplicación de dos dosis del inmunógeno). Además, se demostró que la síntesis de novo de la glicoproteína D a partir del vector viral no replicativo MVA-gDs fue la responsable de la respuesta inmune específica. La inoculación de conejos con virus MVA-gDs (por vía intramuscular o subcutánea) desencadenó una respuesta inmune específica de tipo humoral, observándose la máxima respuesta a los 9 días post dosis de refuerzo. En un ensayo de desafío de conejos con el virus BoHV-1, se observó que la inmunización por vía intramuscular de los animales con virus MVA-gDs disminuyó la excreción del virus de desafío. Finalmente, se demostró que la inmunización de ratones por vía intranasal con virus MVA-gDs adicionados con toxina colérica indujo respuestas inmunes humorales específicas de tipo IgA en muestras de lavados nasales y broncopulmonares y de tipo IgG en muestras de suero. De esta forma, se obtuvo, caracterizó y evaluó un nuevo inmunógeno denominado MVA- gDs que, en el futuro, podría ser evaluado como candidato a vacuna contra el virus BoHV-1 en bovinos.

Delays in construction projects are frequently expensive, since there is usually a construction loan involved which charges interest, management staff dedicated to the project whose costs are time dependent, and ongoing inflation in wage and material prices. Many techniques are used to analyze delays. Some of these methods have inherent weaknesses and should be avoided. This book points out the shortcomings of these faulty methods and explains how a delay analysis should be performed. It then describes specifically how the analysis is done with CPM schedules. A explanation of delays and delay damages, presented in a straightforward, accessible manner, should be useful to public and private owners, construction managers, general contractors, subcontractors, designers, suppliers, and attorneys whose work involves them in the construction industry. The discussion will include subtleties of the process, such as shifts in the critical path, and non-critical delays. The subject of damages is covered in detail, including the major categories of extended field overhead and unabsorbed home office overhead. Likewise, the damages suffered by the owner, either actual or liquidated, are also explained. Finally, a chapter is devoted to managing the risk of delays and time extensions from the viewpoints of the various parties to a construction project. A discussion of early completion schedules and constructive acceleration is also included.

In this new ediion, all chapters are updated to reflect the changes in the construction field since the first edition published over l6 years ago. The 2nd edition includs over 40% more information such as new methods for analyzing delays with examples of the proper approach. The author also includes a new chapter on risk managment which focuses on the delay-related risks of the various parties in a construction project.
